albumin conjugate raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

Naming in this area is inconsistent, and readers should treat product labels with care. In much of the literature and in vendor catalogs, the unqualified term refers to the albumin-binding version, while the version lacking the DAC group appears as modified GRF(1-29), mod GRF(1-29), or the same name with a without-DAC qualifier. Because one abbreviation has been applied to both materials, the only reliable way to identify a sample is to check the stated sequence and the presence of the linker.
CJC-1295 is the name used for a synthetic peptide modeled on growth hormone-releasing hormone, the hypothalamic signal that prompts the pituitary to release growth hormone. The compound was described by a Canadian drug discovery company in the mid-2000s as a long-acting research tool. Two closely related molecules share the name in practice: one carries a drug affinity complex, or DAC, group, and one does not. The distinction matters because the two behave differently in circulation.
The peptide backbone corresponds to GRF(1-29), the first 29 residues of native growth hormone-releasing hormone, which retains most of the receptor-activating activity of the full-length molecule. Four substitutions distinguish the analog from the natural sequence: D-alanine at position 2, glutamine at position 8, alanine at position 15, and leucine at position 27. These changes slow cleavage by dipeptidyl peptidase IV, the enzyme that degrades native hormone in plasma within minutes. The outcome is improved enzymatic stability combined with a still brief residence time when no additional modification is present.
Two forms circulate under the CJC-1295 name, and they differ by a single appended group. The version without a drug affinity complex carries four substitutions along the peptide chain, including a D-alanine near the amino terminus and replacements at three other positions. These changes block the enzyme dipeptidyl peptidase IV and remove a methionine residue that is prone to oxidation. The modified fragment is frequently labeled MOD GRF 1-29. Naming conventions are inconsistent across informal sources, which is a common source of confusion.
The second form adds a maleimide-bearing linker to the lysine at the carboxyl end. This group reacts with cysteine-34 on circulating serum albumin, forming a covalent bond that keeps the peptide in the bloodstream for far longer. ConjuChem developed the molecule as a way to extend the action of a peptide without frequent administration. The albumin attachment is the defining structural feature of the drug affinity complex version. Whether continuous exposure produces effects distinct from shorter pulses remains an unresolved research question.

Batch-to-batch consistency depends on solid-phase peptide synthesis and subsequent purification. Coupling efficiency, resin choice, and cleavage conditions all affect the final profile. Counter-ion content and moisture can shift the apparent mass of a batch. Documentation typically includes a certificate of analysis with chromatograms and spectra. Independent verification by a second laboratory is sometimes requested. Whether a given certificate reflects the actual vial contents depends on chain of custody. Analytical methods themselves carry uncertainty that should be stated alongside results.
Lyophilized material is typically stored at minus twenty degrees Celsius or lower. Keeping the vial dry and protected from light preserves peptide integrity. Repeated freeze-thaw cycles can cause aggregation or loss of activity. Once dissolved, solutions are generally kept at two to eight degrees Celsius. Stability data for reconstituted solutions vary, and long-term behavior is not fully established. Working aliquots reduce the number of times a stock container is opened.
Reverse-phase high-performance liquid chromatography is the standard tool for purity assessment. The technique separates the target peptide from truncated or modified byproducts. Mass spectrometry confirms molecular weight and supports sequence verification. Electrospray ionization and matrix-assisted laser desorption are both used. Amino acid analysis provides an independent check on composition. Purity values are commonly reported as area percentage from the chromatogram. Residual trifluoroacetate and water content are also measured in many quality programs.
The attached maleimide group explains the unusual duration of the DAC version. After injection it reacts with the thiol of cysteine-34 on serum albumin, forming a stable covalent bond. The resulting conjugate is too large for rapid kidney filtration and is shielded from many peptidases. Reported half-lives for this form reach several days, whereas the version without the group is cleared in roughly half an hour. That gap is the main pharmacological difference between the two.
Downstream of growth hormone, the liver and other tissues increase production of insulin-like growth factor 1, a mediator of many growth-promoting effects. Studies have documented elevated levels of both hormones after dosing, and the rise from the long-acting form persists longer than that produced by shorter-acting analogues. What remains unclear is whether sustained elevation of these markers translates into meaningful clinical benefit, and whether prolonged exposure carries risks that short trials could not detect.
CJC-1295 acts at the growth hormone-releasing hormone receptor, a G-protein-coupled receptor found on somatotroph cells in the anterior pituitary. Binding triggers a rise in cyclic AMP and calcium entry, which promotes release of stored growth hormone. Because the peptide mimics the body's own releasing hormone, it amplifies existing secretory pulses rather than driving continuous output. The size of the response therefore depends partly on the subject's own hormonal rhythm and feedback state.

Viral hemorrhagic fevers (VHFs) are a diverse group of infectious diseases characterized by fever and systemic damage to the circulatory system caused by RNA viruses. Viral hemorrhagic fevers cause symptoms ranging from mild to life-threatening, depending on the virus involved, but generally cause internal bleeding that leads to sudden onset of muscle pain, fever, and hypotension. In severe cases, it can cause life-threatening shock and bleeding from internal organs. While some VHFs are generally mild, such as nephropathia epidemica (caused by two species of hantavirus), many are debilitating or lethal without treatment. Outbreaks of VHFs tend to have high case fatality rates and disproportionately affect communities with poor health infrastructure. Therefore, the emergence of VHFs is a growing public health concern. VHFs are often zoonoses, meaning they can be transmitted from wild animals to human populations; for instance, the Lassa arenavirus is spread by mice. Viral hemorrhagic fevers are caused by members of seven families of single-stranded RNA viruses: Arenaviridae, Filoviridae, Flaviviridae, Nairoviridae, Phenuiviridae, Hantaviridae and Peribunyaviridae.
Elsewhere in the province, parts of Spin Boldak District were also struck. Additionally, Taliban forces' facility in Pul-e-Charkhi was also struck by the PAF. Following Pakistan's airstrikes on Kabul and Kandahar, Zabihullah Mujahid stated that the Taliban were ready to negotiate with Pakistan. On 27 February, the PAF carried out airstrikes in parts of Paktika and Laghman provinces, according to local Afghan sources. The airstrike in Laghman targeted the 201 Khalid Ibn Walid Corps, while the one in Paktika struck a house. However, according to some Afghan sources, the Taliban-led Afghan armed forces base in Paktika was the one that was struck by the PAF. Afghan local sources also report an airstrike on the Taliban-led Afghan border brigade in Paktika province. Later the same day, at around noon, the Taliban-led Afghan border brigade in Gardez was also struck by an airstrike carried out by the PAF. An additional airstrike in parts of Nangarhar was also reported by local Afghan sources. The Taliban-led Afghan armed forces base in Khost Province was also struck in the airstrikes that were carried out on 27 February. On the same day, the Taliban-led Afghan Ministry of Defense announced that it had carried out airstrikes in Pakistan targeting military sites in Faizabad, Nowshera, Jamrud, and Abbottabad. The ministry did not provide details about the type of aircraft or equipment allegedly used in the attacks. Pakistani officials, however, stated that attempts to strike in Abbottabad, Swabi, and Nowshera using small drones had been foiled using anti-drone systems.
Female puberty generally occurs between the ages of 9 and 13 and is characterized by ovulation and menstruation; the growth of secondary sex characteristics, such as growth of pubic and underarm hair, breast, uterine and vaginal growth, widening hips and increased height and weight, also occur during puberty. Male puberty sees the further development of the human penis and testicles. The female inner sex organs are the two ovaries, their fallopian tubes, the uterus, and the cervix. At birth there are about 70,000 immature egg cells that degenerate until at puberty there are around 40,000. No more egg cells are produced. Hormones stimulate the beginning of menstruation, and the ongoing menstrual cycles. The female external sex organs are the vulva (labia, clitoris, and vestibule). The male external genitalia include the penis and scrotum that contains the testicles. The testicles are gonads that produce the sperm cells which are ejaculated in semen through the penis. Unlike the egg cells in the female, sperm cells are produced throughout life. Other internal sex organs are the epididymides, vasa deferentia, and some accessory glands. Diseases that affect the reproductive system include polycystic ovary syndrome, a number of disorders of the testicles including testicular torsion, and a number of sexually transmitted infections including syphilis, HIV, chlamydia, HPV and genital warts. Cancer can affect most parts of the reproductive system including the penis, testicles, prostate, ovaries, cervix, vagina, fallopian, uterus and vulva.
=== Cholesterol homeostasis === As a negative post-translational regulator of the low-density lipoprotein receptor (LDLR), PCSK9 plays a major role in cholesterol homeostasis. Upon binding of low-density lipoprotein (LDL) cholesterol to the LDL receptor, the resulting LDLR-LDL complex is internalized. When exposed to the acidic environment within the resulting endosome LDLR adopts a hairpin conformation. This conformational change in turn induces the dissociation of the LDL-LDLR complex, allowing LDLR to be recycled back to the plasma membrane. Binding of PCSK9 to cell surface LDLR (through the LDLR EGF-A domain) also induces LDLR internalization. However, unlike LDL binding, PCSK9 prevents LDLR from undergoing a conformational change. This inhibition redirects LDLR to a lysosome where it is degraded. Thus, PCSK9 lowers cell surface expression of LDLR and thereby decreases metabolism of LDL-particles, which in turn may lead to hypercholesterolemia. PCSK9 also plays an important role in triglyceride-rich apoB lipoprotein production in small intestine and postprandial lipemia.

The city has a rich sporting heritage as the home to two historically significant football teams: Juventus FC (founded in 1897) and Torino FC (founded in 1906). Juventus has the larger fan base, especially all over Italy and worldwide, while Torino enjoys a greater support in the city itself. The two clubs contest the oldest derby in Italy, the Derby della Mole. Juventus is Italy's most successful football club and one of the most successful in the world. It ranks joint twelfth in the list of the world's clubs with the most official international titles (sixth between European clubs), and was the first in association football history—remaining the only one in the world (as of 2022, after the first UEFA Europa Conference League Final)—to have won all possible official continental competitions and the world title. Juventus' owned ground, the Juventus Stadium, was inaugurated in 2011 and hosted the 2014 UEFA Europa League final, the 2021 Nations League finals and the 2022 Women's Champions League final. This was the first time the city hosted a seasonal UEFA club competition's single-match final. Juventus' female team has won, among others, five straight Serie A titles since its inception in 2017, becoming one of the most winning teams in the country. Torino FC was founded by the union of one of the oldest football teams in Turin, Football Club Torinese (founded in 1894), with breakaways from Juventus and was the most successful team, called "Grande Torino", in the Serie A during the 1940s.

=== Hacktivism === Politically motivated hacktivism involves the subversive use of computers and computer networks to promote an agenda, and can potentially extend to attacks, theft and virtual sabotage that could be seen as cyberwarfare – or mistaken for it. Hacktivists use their knowledge and software tools to gain unauthorized access to computer systems they seek to manipulate or damage not for material gain or to cause widespread destruction, but to draw attention to their cause through well-publicized disruptions of select targets. Anonymous and other hacktivist groups are often portrayed in the media as cyber-terrorists, wreaking havoc by hacking websites, posting sensitive information about their victims, and threatening further attacks if their demands are not met. However, hacktivism is more than that. Actors are politically motivated to change the world, through the use of fundamentalism. Groups like Anonymous, however, have divided opinion with their anarchic methods.
=== Pregnancy === In women with known hypothyroidism who become pregnant, it is recommended that serum TSH levels are closely monitored. Levothyroxine should be used to keep TSH levels within the normal range for that trimester. The first-trimester normal range is below 2.5 mIU/L and the second and third trimesters normal range is below 3.0 mIU/L. Measurement of free T4 in pregnancy is not recommended due to changes in levels of serum protein binding. Similarly to TSH, the thyroxine results should be interpreted according to the appropriate reference range for that stage of pregnancy. The levothyroxine dose often needs to be increased after pregnancy is confirmed, although this is based on limited evidence and some recommend that it is not always required; decisions may need to based on TSH levels. Women with anti-TPO antibodies who are trying to become pregnant (naturally or by assisted means) may require thyroid hormone supplementation even if the TSH level is normal. This is particularly true if they have had previous miscarriages or have been hypothyroid in the past. Supplementary levothyroxine may reduce the risk of preterm birth and possibly miscarriage. The recommendation is stronger in pregnant women with subclinical hypothyroidism (defined as TSH 2.5–10 mIU/L) who are anti-TPO positive, in view of the risk of overt hypothyroidism. If a decision is made not to treat, close monitoring of the thyroid function (every 4 weeks in the first 20 weeks of pregnancy) is recommended.
=== Thirds, fourths === The term "minute" usually means 1⁄60 of an hour, coming from "a minute division of an hour". The term "second" comes from "the second minute division of an hour", as it is 1⁄60 of a minute, or 1⁄60 of 1⁄60 of an hour. While usually sub-second units are represented with SI prefixes on the second (e.g. milliseconds), this system can be extrapolated further, such that a "third" would mean 1⁄60 of a second (16.7 milliseconds), and a "fourth" would mean 1⁄60 of a third (278 microseconds), etc. These units are occasionally used in astronomy to denote angles.

No. It is a laboratory-made analog of growth hormone-releasing hormone. The natural hormone is a 44-residue peptide, while the analog is built on a shorter 29-residue fragment.
The original material included a linker that binds serum albumin. A later variant removed that linker to reduce the duration of action. Catalogs and papers did not consistently adopt separate names, so both remain widely labeled with the same term.
The amino acid sequence and the presence or absence of the linker group. Mass measured by spectrometry gives an independent check that separates the two forms.
The powder is normally held at minus twenty degrees Celsius or below. Light and moisture exposure should be minimized. Repeated warming and cooling cycles are avoided.
